摘要:【目的】明确山东省济宁、泰安、潍坊3市核桃细菌性黑斑病的病原,为病害防治提供依据。【方法】采用实地踏查法,调查3市核桃园黑斑病发生情况并采集样品,采用组织分离法对病原菌进行分离,人工接种叶片进行病菌的致病性测定,最后利用16S rDNA序列比对和系统进化树分析进行病原菌鉴定,同时也应用gyrB基因系统进化准确鉴定黄单胞菌(Xanthomonas spp.)。【结果】结果表明,引起山东省3市核桃黑斑病的病原菌主要有成团泛菌(Pantoea agglomerans)、变黄假单胞菌(Pseudomonas flavescens)以及栖树黄单胞菌(X.arboricola),其中变黄假单胞菌引起核桃黑斑病是国内首次报导。【结论】成团泛菌、变黄假单胞菌、栖树黄单胞菌是山东省3市核桃黑斑病的主要病原菌。
【Objective】To clarify the pathogen of bacterial black spot disease in walnut trees in Weifang,Jining,and Tai'an,Shandong Province,and provide a basis for disease control.【Method】Employing on-site field survey methodology to investigate the incidence of black spot disease in walnut orchards across three cities and collected representative samples.Pathogenic microorganisms were isolated using tissue separation techniques,followed by pathogenicity assessments through artificial inoculation on leaves.The identification of the pathogen was subsequently conducted through 16S rDNA sequence alignment and systematic phylogenetic tree analysis and the gyrB gene was also applied to accurately identify Xanthomonas spp.【Result】The pathogens causing walnut black spot disease in three places in Shandong Province were Pantoea agglomerans,Pseudomonas flavescens and Xanthomonas arboricola,of which Pseudomonas flavescens was first reported in China.【Conclusion】Pantoea agglomerans,Pseudomonas flavescens and X.arboricola are identified as the main pathogen of walnut black spot disease in three cities of Shandong province.
