摘要:以大百合中层鳞茎作为外植体,研究了培养基、激素配比、蔗糖浓度和光照强度对大百合鳞茎诱导出芽的影响。结果表明:大百合鳞茎诱导的最适培养基为MS作为基本培养基,前4周采用暗培养,以MS+6-BA 5.0 mg/L+NAA0.1 mg/L+蔗糖45 g/L为培养基诱导愈伤组织,之后采用光培养,以MS+6-BA 0.5 mg/L+IBA 0.5 mg/L+蔗糖15 g/L为培养基诱导愈伤组织分化出芽,可使愈伤组织诱导率达到88.89%,出芽率达到93.82%,平均出芽数达到12.3,能够诱导产生大量材料,有利于大百合组培快繁。
The middle-layer bulbs of Cardiocrinum giganteum were taken as explants,and the effects of culture medium,hormone combination,sucrose concentration and illumination intensity on the bud induction from bulbs of C. giganteum were studied. The results showed that the optimum basic culture medium for bud induction from bulbs of C. giganteum was MS. MS+ 6-BA5.0 mg/L+ NAA 0.1 mg/L+ 45 g/L sucrose was the optimum medium for callus induction under dark condition in the first four weeks,and MS+ 6-BA 0.5 mg/L+ IBA 0.5 mg/L+ 15 g/L sucrose was the optimum medium for bud induction under illumination condition after that. Through above-mentioned culture,the callus induction rate reached 88.89%,the budding ratio reached93.82%,and the average number of buds per explant was 12.3.
