摘要:用随机扩增多态性DNA (RAPD)技术对核桃属内 9个种及近缘属的 2个种进行基因组DNA多态性分析 ,共选用 2 0个 10bp随机引物扩增出 311个DNA片段 ,其中铁核桃J .sigillata、甘肃枫杨P .macroptera显示出自身特征性标记 ,利用这些片段进行种间遗传关系分析 ,根据UPGMA方法构建聚类树状图。研究结果表明 :铁核桃为核桃属中一个独立种 ;核桃属内组间和种间亲缘关系与经典分类学的结果完全一致 ;RAPD对核桃属与近缘属的分析结果有待进一步研究证实。
Random amplified polymorphic DNA(RAPD)technique was used to analyze the genetic relationship of 11 species of Juglans and its relatives.Twenty decanucleotide arbitrary primers were selectol and 311 DNA fragments were generated,these fragments were used in the analysis of the relationship of the 11 species.The dendrogram was constructed by using UPGMA methods based on their genetic distance,giving the taxonomic position of these species.Specific markers were found in J.sigillata ,this indicates that it is a independent species.The availability of RAPD in the analysis of the genetic relationship at the genus level needs further study.
