摘要:以湖南地区13个南天竹种源为研究材料,采用常规观测和现代生物技术相结合的研究方法,对来自不同地区的南天竹形态特征和遗传多样性进行了系统研究.结果表明,南天竹资源丰富,变异类型多;各性状在不同地理种源之间变异系数较大,平均变异达到了29.956%;各性状变异系数的大小次序为叶>树体>花>果.利用ISSR分子标记对南天竹种源进行遗传多样性分析,从100条随机引物中筛选出10条重复性好、条带特异、多态性明显的引物,平均每条引物可扩增出4.8条多态带,多态率为85.7%.13个种源ISSR标记聚类为4类,各类都有明显的分布地域.邵东南天竹和宜章南天竹之间的遗传距离最小,相应的遗传一致度最大;张家界南天竹与衡山南天竹之间的遗传距离最大,相应的遗传一致度最小.南天竹形态标记和ISSR标记聚类结果之间的相关性不高,相关系数仅为0.213.
Thirteen sources of Nandina domestica from Hunan of China were taken as the materials, of which routine observetion and morden molecular technology were used to detect the morphological and genetic diversity. The results of morphological research showed that genetic diversity was much abundant in Nandina domestica in Hunan. The average variation coefficient(CV)was 29. 956% among the 25 morphological characters. The order of variation coefficient of phenotypic were leaf,body size, flower and fruit. The improved ISSR-PCR reaction system is established for the genetic diversity analysis of 13 populations of Nandina domestica in Hunan Province, and 10 primers characterized by the good repetition, special bands and distinct polymorphism were selected from 100 random ISSR primers. Each selected primer averagely yielded 4.8 potymorphic bands,the polymorphic rate reaching 85.7%. According to UPGMA clustering results and ISSR analysis and their geographical origins characters, 13 sources from Hunan were classified into 4 clusters. All kinds had differention area. The results of genetic distance(Nei' s)and genetic similarity coefficient showed: the genetic distance was the least between the Shaodong Nandina domestica and the Yizhang Nandina domestica,and the corresponding genetic similarity coefficient was the biggest. But the Nei' s genetic distance was the farthest between the Zhangjiajie Nandina domestica and the Hengshan Nandina domestica, and the corresponding genetic similarity coefficient was the smallest. The relevant coefficient between the morphological mark and the molecular mark was 0. 213 ,both correlation were not higher.
