摘要:为研究西藏拉萨引种的4种竹子在自然低温过程中的耐受生理学特性和耐寒性差异,以引种于西藏拉萨近2 a的西藏箭竹、错那箭竹、紫竹和金镶玉为试验材料,对4个竹种在自然低温下的丙二醛(MDA)含量、过氧化物酶活性(POD)活性、可溶性蛋白含量、可溶性糖含量和脯氨酸含量进行测定,通过相关分析、冗余分析(RDA)和聚类分析,探究自然低温环境下4个竹种的生理响应和耐寒性差异,并采用隶属函数法综合评价各竹种的耐寒性。结果显示:随气温的变化,4种竹子叶片中丙二醛(MDA)含量、过氧化物酶活性(POD)活性、可溶性蛋白、可溶性糖含量和脯氨酸含量总体呈先升高后降低的变化趋势,在气温降低后各指标含量达较高水平(紫竹和金镶玉可溶性蛋白除外,因其含量在低温驯化后显著降低);耐寒性强的西藏箭竹和错那箭竹保持较低水平的MDA含量(与紫竹相比,MDA含量平均低出38.37%~68.33%),可溶性蛋白、脯氨酸含量则保持较高水平(与紫竹和金镶玉相比,平均高出7.19%~81.31%、57.01%~133.14%);隶属函数法综合评价4种竹子耐寒性,表现为错那箭竹>西藏箭竹>金镶玉>紫竹。研究结果表明,在拉萨进行绿化竹子引种时,应优先考虑当地的错那箭竹、西藏箭竹等耐寒性较强的高山竹种。
To study the physiological characteristics and cold tolerance differences of four woody bamboos introduced in Lasa,Xizang,China under natural low temperature,physiologic indicators of these four species including Fargesia macclureana,F.grossa,Phyllostachys nigra and P.aureosulcata f.spectabilis,were observed.The malondialdehyde(MDA)content,peroxidase activity(POD)activity,soluble protein content,Soluble sugar content and proline content were measured,and the physiological responses and cold tolerance differences of these four species were investigated using correlation analysis,redundancy analysis(RDA)and cluster analysis,and the membership function method was employed to comprehensively evaluate the cold tolerance of four bamboo species.The results showed that the MDA content,POD activity,soluble protein content,soluble sugar content,and proline content in the four bamboo leaves increased at first and then decreased with temperature changes,and reached a higher level after natural low temperature acclimation(except the soluble protein of P.nigra and P.aureosulcata f.spectabilis,the content was significantly reduced after low temperature acclimation).The cold-tolerant F.macclureana and F.grossa maintained a low level of MDA content(compared with P.nigra,the MDA content was lower on average 38.37%~68.33%),while the soluble protein content and proline content maintained a high level(compared with P.nigra and P.aureosulcata f.spectabilis,the soluble protein content was higher on average 7.19%~81.31%,and the proline content was higher on average 57.01%~133.14%),which played a major role in the cold tolerance process.Using the membership function method to comprehensively evaluate the results,it was concluded that the cold tolerance of four species showed a decreasing order of F.grossa>F.macclureana>P.aureosulcata f.spectabilis>P.nigra.Altogether,priority should be given to native alpine bamboo species with strong cold resistance,such as F.grossa and F.macclureana,when introducing ornamental bamboos for landscaping in Lasa.
