摘要:以厚朴(Magnolia officinalis Rehd.et Wils)叶片为材料,采用SDS-CTAB结合法、高盐低pH值法、CTAB法、SDS法等4种方法分别对2种方法保存的样品及对照样(鲜叶)进行了基因组DNA提取效果的比较,并进行ISSR-PCR检测。结果表明:4种方法均能从厚朴叶片中获得高质量的DNA,其中以SDS-CTAB结合法所得的DNA纯度最高,高盐低pH值法次之,CTAB法提取的厚朴叶片DNA纯度最低;高浓度NaAc的引入及相应的冻融,可以提高所得厚朴基因组DNA的纯度;硅胶干燥和-70℃超低温保存的样品与鲜叶相比,所提DNA的质量无明显差异,均能满足PCR扩增的要求。
Experiment was conducted with leaves of Magnolia officinalis stored at-70℃,desiccated by silica gel and 4 methods,including simple CTAB method,improved SDS method,modified SDS-CTAB method and high salt and low pH method,which were used for total DNA extraction for studying their effects on DNA extraction.The results show that DNA can be obtained with all the methods;and of them,the modified SDS-CTAB method extracted the best purity of DNA,the high salt method was the next and the simple CTAB was the lowest.The purity of DNA can be raised by high concentration sodium acetate and freeze-thaw.The purity and yields of DNA extracted from the leaves preserved in silica gel or-70℃ were not significantly different from those of fresh leaves,and they could meet the requirement of PCR amplification.
