摘要:【目的】旨在了解雄全同株性别类型‘龙岩野柿1号’雄花和两性花花芽转录组中的SSR位点及序列特征,为SSR分子标记开发提供理论依据。【方法】基于雄全同株种质‘龙岩野柿1号’花芽分化的两个形态学关键时期的雄花和两性花花芽转录组序列,运用MISA软件的默认参数即单碱基类型SSR重复数≥10、双碱基类型重复数≥6和三、四、五、六碱基类型重复数≥5对转录组获得的unigenes序列中的SSR位点进行搜索,分析SSR位点的序列特征。【结果】对‘龙岩野柿1号’转录组的82910条unigenes进行搜索,共在25349条unigenes上获得了38751条SSR序列,发生频率为30.57%,出现频率为46.74%,平均每2.94 kb会出现1个SSR序列;双碱基重复类型SSR位点数目最多,占总SSR数的50.59%,单碱基重复类型SSR数目次之,占比24.59%;单碱基重复至三碱基重复类型的优势重复基元分别为A/T、AG/CT和AAG/CTT,分别占比22.8%、35.15%和7.77%,四碱基重复至六碱基重复类型的优势重复基元分别为AAAG/CTTT、AAAAG/CTTTT和AGGGCG/CCCTCG,占比分别为0.45%、0.09%和0.05%;SSR位点的基元重复次数以5~20为主,序列长度变化范围较大,为10~94 bp,共有11731个SSR位点的序列长度≥20 bp,占比为30.27%。【结论】‘龙岩野柿1号’花芽的转录组中SSR位点丰富,具有较高的多态性,在柿属植物性别类型相关的分子标记开发、分子标记辅助育种和遗传多样性分析中具有较大的应用潜力。
【Objective】The aim of this study was to analyze the SSR loci in the transcriptome of male and hermaphroditic floral buds of Diospyros kaki‘Longyanyeshi 1’,to provide a basis for the development of SSR molecular markers.【Method】The SSR loci were searched and characterized using the default parameters of MISA software based on the transcriptome sequences of male and bisexual flower buds during the two morphologically critical periods of flower bud differentiation in‘Longyanyeshi 1’.【Result】By searching the 82910 unigenes in the transcriptome of‘Longyanyeshi 1’,a total of 38751 SSR sequences were obtained from 25349 unigenes,with an occurrence frequency of 30.57%and a frequency of 46.74%,with an average length of 2.94 kb between two SSR loci.The number of dinucleotide repeat SSR sites was the largest,accounting for 50.59%of the total SSRs,followed by the mononucleotide repeat type,accounting for 24.59%.The dominant repeat motifs of mononucleotide,dinucleotide and trinucleotide repeat types were A/T,AG/CT and AAG/CTT,accounting for 22.8%,35.15%and 7.77%,respectively.The dominant repeat motifs of tetranucleotide,pentanucleotide,hexanucleotide repeat types were AAAG/CTTT,AAAAG/CTTTT and AGGGCG/CCCTCG,accounting for 0.45%,0.09%and 0.05%,respectively.The motif repetition times of SSR loci were mainly from 5 to 20,and the sequence length varied from 10 to 94 bp.There were 11731 SSR loci with a sequence length≥20 bp,accounting for 30.27%of the total SSRs.【Conclusion】The transcriptome of‘Longyanyeshi 1’is rich in SSR loci with high polymorphism and has great application potential in the development of molecular markers related to sex types,molecular marker-assisted breeding and genetic diversity analysis.
