摘要:通过常规组织分离法对采自陕西省陕南和关中的核桃(Juglans regia)炭疽病样品进行了分离,采用形态学、培养特性并结合核糖体内转录间隔子(rDNA-ITS)、β-微管蛋白(TUB2)、钙调蛋白(CAL)、谷氨酰胺合成酶(GS)和3-磷酸甘油醛脱氢酶(GAPDH)的多基因系统发育分析方法以及致病性测定,明确了引起陕西省核桃炭疽病病原菌的种类。结果显示:12个代表性菌株被鉴定为胶孢刺盘孢(Colletotrichum gloeosporioides),4个代表性菌株被鉴定为尖孢刺盘孢复合种(C.acutatum)中的C.sp.,C.sp.与古代稀刺盘孢(C.godetiae)在系统进化关系上最接近,自展支持率为91%。致病性测定显示,胶孢刺盘孢和C.sp.能够侵染核桃果实并产生典型的炭疽病症状。
In order to determine the Colletotrichum species caused by walnut anthracnose in Shaanxi Province, the samples with symptom of walnut anthracnose were collected from southern Shaanxi and Guanzhong areas. Isolates were recovered from the samples by tissue isolation method. Isolates were identified as Colletotrichum species based on morphological characteristics, cultural characteristics and multigene phylogenic analysis of the ribosomal internal transcribed spacer(rDNA-ITS), β-tubulin(TUB2), calmodulin(CAL), glutamine synthetase(GS) and glyceraldehyde-3-phosphate dehydrogenase(GAPDH) genes. Pathogenicity tests on walnut fruit were conducted using two representative isolates from each species. The results showed that 12 representative isolates were identified as C. gloeosporioides, and 4 representative isolates were identified as Colletotrichum sp. belonging to the C. acutatum complex. The pathogen Colletotrichum sp. was closest to C. godetiae in terms of phylogenetic evolution, with 91% supporting rate. Pathogenicity tests indicated that C. gloeosporioides and Colletotrichum sp. could infect walnut fruits and cause typical symptoms of walnut anthracnose.
