摘要:采用邻苯三酚自氧化法、DPPH法、Fenton反应体系、FRAP分析法及对过氧化氢损伤细胞保护作用方法对白蜡虫Ericerus pela Cavannes雌成虫多糖、蝗虫多糖、喙尾琵甲多糖、美洲大蠊多糖及家蚕蛹多糖进行体外抗氧化研究,评价昆虫多糖总还原能力,对超氧阴离子自由基(O2-·)、DPPH·、羟自由基(·OH)的清除作用及损伤细胞存活率影响。试验结果表明,5种多糖对几种自由基均有不同程度的清除作用,且均呈现一定的量效关系;但对不同的氧化体系呈现不同的清除活性,白蜡虫多糖的总还原能力最强;白蜡虫、喙尾琵甲、美洲大蠊及蝗虫多糖对DPPH的清除率相对较明显;在羟自由基的清除能力上蚕蛹、喙尾琵甲及美洲大蠊多糖相对较强;各昆虫多糖对超氧阴离子清除能力不明显。5种多糖对过氧化氢损伤细胞有一定保护作用,在不同浓度范围内能增加细胞存活率。研究表明昆虫多糖具有一定抗氧化活性,可成为天然抗氧化物的来源。
Antioxidant activities of five insect polysaccharides, extracted from white wax female scale, cockroach, black beetle, locust and silkworm pupa, were evaluated in vitro, which including DPPH radical scavenging assay, superoxide radical scavenging assay, ferric reducing antioxidant powder assay (FRAP) , hydroxyl radical scavenging assay, and proliferation effect of SH -SY5Y cells induced by hydrogen peroxide. The polysaccharides were found to have different levels of antioxidant activity in the systems tested. The experimental data indicated that the antioxidant activities of five polysaccharides were different. The results showed that FRAP value of polysaccharide extracted from white wax female scale was higher than others'. The polysaccharide extracted from silkworm pupa had the lowest scavenging effect on DPPH in five polysaccharides, and the hydroxyl radical scavenging activity of polysaccharide from locust was lowest. The scavenging superoxidant radical capacities of all samples were not obvious. The five insect polysaccharides could increase proliferation of cells damaged by hydrogen peroxide in different concentration. The results showed that five insect polysaceharides had antioxidant activities and could be explored as natural new potential antioxidant.
