摘要:以橡椀壳粉为原料,采用工业纤维素复合酶辅助提取橡椀单宁,在单因素试验基础上通过响应面法优化提取工艺,进一步研究水酶法提取过程中对应的动力学和热力学模型。研究结果表明:橡椀单宁的最佳提取溶液pH值5.0、1.00 g橡椀壳粉、料液比1∶50(g∶mL)、提取时间27.7 min、提取温度55℃、酶用量为橡椀壳粉质量的1.9%,此条件下单宁提取率为97.56%。在该优化工艺条件下,原料质量放大至90 g时,单宁提取率仍达到97.3%,且提取物中单宁质量分数为55.0%、总多糖质量分数为5.16%、总黄酮质量分数为2.95%。动力学和热力学研究表明:单宁提取过程符合Fick第二定律,293~353 K范围内,速率常数(k)为0.1220~0.2596 min^(-1),表观活化能(E_(a))为10.683 kJ/mol,指前因子(A)为9.6,提取过程的焓变(ΔH)为28.584 kJ/mol、熵变(ΔS)为110.576 J/(mol·K)、吉布斯自由能变(ΔG)为-8.652~-4.074 kJ/mol,表明该提取过程可自发进行,升高温度有利于橡椀单宁的提取。
Using valonia shell powder as raw material,industrial cellulose composite enzymes were used to assist in the extraction of valonia tannins.Based on single factor experiments,the extraction process were optimized by response surface methodology,and the corresponding kinetics and thermodynamic models during the aqueous enzymatic extraction process were further studied.The results showed that the optimal extraction conditions for valonia tannins was pH of 5.0,valonia shell powder of 1.00 g,solidliquid ratio of 1∶50(g∶mL),extraction time of 27.7 min,extraction temperature of 55℃,the enzyme dosage was 1.9%of the weight of valonia shell powder.Under these conditions,the tannin extraction rate was 97.56%.Under these optimized conditions,when the mass of raw materials was magnified to 90 g,the extraction rate of tannin still reached 97.3%.The tannin content in the extract was 55.0%,the total polysaccharide mass fraction was 5.16%,and the total flavonoid mass fraction was 2.95%.In addition,the kinetic and thermodynamic studies showed that the tannin extraction process conformed to the Fick'second law.In the range of 293-353 K,the rate constant(k)was 0.1220-0.2596 min^(-1),the apparent activation energy(E_(a))was 10.683 kJ/mol,and the pre-exponential factor(A)was 9.6.During the extraction process,the enthalpy change(ΔH)was 28.584 kJ/mol,the entropy change(ΔS)was 110.576 J/(mol·K),Gibbs free energy change(ΔG)was-8.652--4.074 kJ/mol,indicating that the extraction process could proceed spontaneously and increasing the temperature was beneficial for the extraction of valonia tannins.
