摘要:【目的】探究蒸汽爆破(汽爆)处理对杜仲叶片水提液抗氧化能力和香气组分的影响。【方法】以紫色杜仲品种‘华仲12’的叶片为试验材料,汽爆(压强0.45 MPa,稳压时间300 s)前设杀青后揉捻(A)、杀青(B)、萎凋后揉捻(C)和仅萎凋(D)4个处理,并设仅萎凋不汽爆对照(CK),分别测定各处理叶片水提取液的1,1-二苯基-2-苦基肼(DPPH)自由基、2,2’-连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸)二铵盐(ABTS^(+))自由基、羟自由基、超氧阴离子清除能力及总抗氧化能力(FRAP值),用HPLC测定原儿茶酸、绿原酸、京尼平苷酸、儿茶素、没食子酸甲酯、松脂醇二葡萄糖苷、车叶草苷含量,并分析上述所有指标的相关性,再用顶空固相微萃取-气相色谱-质谱联用法(HS-SPME-GC-MS)检测挥发性物质成分,比较汽爆表现最佳处理与对照的成分差异。【结果】杜仲叶片经杀青(B)处理再汽爆的效果最好,其水提取液活性成分含量和体外抗氧化能力较高,总多酚和总黄酮含量最高,分别为401.59和62.65 mg/g,较对照分别提高了62%和370%;该处理7种活性成分含量是对照的1.68~21.92倍。杜仲叶提取物质量浓度为0.2~6.0,2.0~12.0,1.25~40.00,1.25~40.00 g/L时,B处理提取物清除DPPH、ABTS^(+)、羟自由基、超氧阴离子的IC_(50)值分别为0.814,1.956,0.977和16.029 g/L,其FRAP值在提取物质量浓度为0.30 g/L时达最高(1.126 mmol/g),较对照提高了40.4%。相关性分析表明,车叶草苷、松脂醇二葡萄糖苷、京尼平苷酸、绿原酸、儿茶素、没食子酸甲酯、原儿茶酸和总黄酮含量与DPPH自由基、ABTS^(+)自由基、超氧阴离子清除能力显著相关。与对照相比,汽爆后表现最佳的B处理多出17种特有香气的挥发性化合物,包括特有香气物质苯甲醇(相对含量7.53%)、4-(2,6,6-三甲基-1,3-环己二烯-1-基)-3-丁烯-2-酮(相对含量0.68%)、α-异甲基紫罗酮(相对含量0.30%)和β-环柠檬醛(相对含量2.64%)。【结论】汽爆处理显著提高了紫色杜仲叶片水提液活性成分含量和抗氧化能力,丰富了挥发性化合物组成。
【Objective】The study aimed to investigate the effect of steam explosion(SE)treatment on the antioxidant capacity and aroma components of aqueous extract of Eucommia ulmoides leaves.【Me-thod】E.ulmoides‘Huazhong 12’leaves were pretreated before SE(pressure 0.45 MPa,retention time 300 s)using four different approaches:blanching and rolling(A),blanching only(B),withering and rolling(C),withering only(D).Additionally,a control group(CK)underwent only withering without SE treatment.The radical scavenging capacity of the treatments was measured for 1,1-Diphenyl-2-picrylhydrazyl(DPPH)radical,2,2’-Azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)diammonium(salt ABTS^(+))radical,hydroxyl radical,superoxide anion scavenging,and ferric reducing antioxidant power(FRAP).HPLC was used to measure protocatechuic acid,chlorogenic acid,geniposidic acid,catechins,methyl gallate,pinoresinol diglucoside,and asperuloside.The correlation of all these indexes was then analyzed.Volatile substance components were detected using headspace solid-phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS),and the differences in these components between the treatments before and after steam explosion were compared.【Result】The effect of blanched(B)‘Huazhong 12’leaves treated by SE was the best.The highest total polyphenol and total flavonoid contents reached 401.59 and 62.65 mg/g,increased by 62%and 370%compared to the control,respectively.The contents of 7 active ingredients were 1.68-21.92 times of the control.The IC_(50)values for B-treated extracts of the DPPH,ABTS^(+),hydroxyl radical,and superoxide anion scavenging were 0.814,1.956,0.977,and 16.029 g/L,respectively,and the FRAP value was 1.126 mmol/g at 0.30 g/L,40.4%increase compared to the control.Correlation analysis showed that carnosine,turpentine diglucoside,kynurenine glycosides,chlorogenic acid,catechin,methyl gallate,protocatechuic acid,and total flavonoids were significantly and positively correlated with DPPH radicals scavenging,ABTS^(+)radicals scavenging,superoxide anion scavenging capacity.Compared with the control,the best-performing treatment B steam-exploded aqueous extract of E.ulmoides produced 17 more volatile compounds of the characteristic aroma.The distinctive aroma substances were benzyl alcohol(7.53%),4-(2,6,6-trimethyl-1,3-cyclohexadien-1-yl)-3-buten-2-one(0.68%),α-isomethyl viologen(0.30%),andβ-cyclocitral(2.64%).【Conclusion】The SE treatment significantly increased the content of active ingredients and antioxidant capacity of the aqueous extract of purple E.ulmoides leaves,and enriched the composition of volatile compounds.
