摘要:为了筛选产没食子酸脱羧酶的菌种并对其相应的酶活性质进行研究,文中首先利用底物诱导对可以产没食子酸脱羧酶的不同菌种进行了筛选,初步研究了没食子酸脱羧酶的专一性及影响其酶活的因素,由结果可知:待筛选的5株菌种均能降解没食子酸生成焦性没食子酸,其降解效果由强到弱为弗式柠檬酸杆菌(Citrobacter freandii)CICC 22001、阴沟肠杆菌(Enterobacter cloacae)CICC 10017、肠杆菌属(Enterobacter spp.)CICC 23468、阴沟肠杆菌(Enterobacter cloacae)CICC 20772、阴沟肠杆菌(Enterobacter cloacae)CICC 20804;没食子酸脱羧酶(GAD)对没食子酸具有较强的专一性,在粗酶液5.0 m L,没食子酸0.4 mg/m L,发酵温度35℃,磷酸盐缓冲溶液pH=6.0对其酶活的促进作用最强,而金属离子的促进作用强弱为Mg^(2+)>Sn^(2+)>Cu^(2+)>Zn^(2+)>K+>Ca^(2+)>Fe^(2+),添加剂的抑制作用强弱为EDTA>SDS>Triton X-100>Tween 20。
In order to screen the strains to produce the gallic acid decarbolxylase and study of their enzymic activities, the different bacterial strains with the abilities of degrading gallic acid were screened by the substrate induction test, and the enzymatic specificity and properties were studied preliminarily. The results show that all of 5 screening strains can degrade gallic acid to pyrogallol, and their effect from strong to weak is Citrobacterfreandii CICC 22001, Enterobacter cloacae CICC 10017, Enterobacter spp. CICC 23468, Enterobacter cloacae CICC 20772, Enterobacter cloacae CICC 20804; GAD has strong enzymatic specificity in degrading gallic acid. Phosphate buffer solution has the strongest promoting effect at 5. 0 mL GAD, 0. 4 mg/mL gallic acid, 35℃ fermentation temperature, and pH 6.0. The effect of metal ions on GAD from strong to weak is Mg2+ 〉 Sn2 + 〉 Cu2+ 〉 Zn2+ 〉 K+ 〉 Ca2+ 〉 Fe2+ , and the effect of the inhibition additives from strong to weak is EDTA 〉 SDS 〉 Triton X-100 〉 Tween 20.
