摘要:为解决市售橡椀栲胶纯度低、杂质多、生物活性不佳等问题,以工厂热水粗提橡椀栲胶为原料,经不同体积分数(70%~95%)乙醇醇沉工艺和不同截留分子质量(3、5、10、20、100 ku)超滤膜分离纯化工艺,得到纯度较高的橡椀单宁。优化了纯化工艺条件,分析了超滤膜纯化过程的动力学参数,并测定了纯化后橡椀单宁的抗氧化活性。研究结果表明:橡椀栲胶经85%乙醇醇沉后,再经10 ku超滤膜在料液质量浓度10 g/L、温度20℃、压力0.35 MPa条件下超滤纯化,最终透过液中单宁纯度由60.8%提升至75.40%,单宁收率为73.28%;膜分离动力学模型得出超滤传质系数(k)为6.59 mm/h,膜面平衡质量浓度(C_(g))为12.14 g/L,操作压力(Δp)与膜通量(J_(v))的关系为J_(v)=Δp/(0.0268+0.0292Δp)。抗氧化活性试验结果表明:不同质量浓度的橡椀单宁对DPPH·、·ABTS^(+)、O_(2)^(-)·、·OH这4种自由基均具有较高的清除能力,半数抑制质量浓度(IC_(50))分别为36.89、39.37、31.58和36.24 mg/L。相同质量浓度下,橡椀单宁具有接近或超过Vc的抗氧化能力,其中高质量浓度橡椀单宁对·OH的清除能力优于Vc。
To solve the problems of low purity,high impurities,and poor biological activity of commercially available valonia tannin extract,factory hot water crude extraction of valonia tannin extract was used as raw material.Through different volume fractions(70%-95%)ethanol alcohol precipitation processes and ultrafiltration membrane separation and purification processes with different cut-off molecular weights(3,5,10,20,100 ku),high-purity valonia tannin was obtained.The purification process conditions were optimized,the kinetic parameters of ultrafiltration membrane purification process were analyzed,and the antioxidant activity of purified valonia tannin was determined.The research results showed that after 85% ethanol precipitation,valonia extract was purified by ultrafiltration using a 10 ku ultrafiltration membrane at a mass concentration of 10 g/L,temperature of 20℃,and pressure of 0.35 MPa.The final tannin purity in the permeate increased from 60.8% to 75.40%,and the tannin yield was 73.28%.The membrane separation kinetics model shows that the ultrafiltration mass transfer coefficient(k)is 6.59 mm/h,the membrane surface equilibrium mass concentration(C g)is 12.14 g/L,and the relationship between operating pressure(Δp)and membrane flux(J_(v))is J_(v)=Δp/(0.0268+0.0292Δp).The antioxidant activity test showed that different concentrations of valonia tannin had high scavenging ability against four free radicals,DPPH·,·ABTS^(+),O_(2)^(-)·and·OH,with half maximal inhibitory concentrations(IC_(50))of 36.89,39.37,31.58,and 36.24 mg/L,respectively.At the same concentration,valonia tannin has antioxidant capacity close to or exceeding that of Vc,and high concentration of valonia tannin has better ability to remove·OH than Vc.
