摘要:从40条RAPD随机引物中筛选了13条和9条,分别对11种倍蚜和角倍蚜4个地理种群的DNA进行PCR扩增和分析。结果表明:倍蚜的遗传距离在不同属之间为0.4828±0.1708,不同种之间为0.2520±0.1780,不同亚种之间为0.1472±0.0764,聚类分析反映了倍蚜属间、种间的亲缘关系及其远近程度,与形态分类结果基本一致;圆角倍蚜属与其他3个属的差异明显,可能是倍蚜中较早分化的类群。角倍蚜不同种群间的遗传距离为0.0759±0.0302,种群间具有丰富的DNA序列多态性并出现了一定程度的遗传分化,造成这种分化的原因可能是地理上的隔离。
The RAPD technique was used to determine phylogenetic relationships of Chinese gallnut aphids and genetic differentiation in four populations of Schlechtendalia chirtensis, a major Chinese gallnut aphid. 13 primers and 9 primers were selected from 40 primers and used to amplify their genome DNA. The genetic distances between genera, species and subspecies are 0.482 8 ± 0. 170 8,0.252 0 ± 0. 178 0,0. 147 2 ± 0.076 4, respectively. Dendrogram based on Nei's genetic distance displayed the phylogenetic relationships of different taxa and almost coincided with morphological taxonomy. Our result suggested that there is a distinct difference between the Nurudea and other three genera. It may be implied that the Nurudea was the first differentiation group among Chinese gall aphids. The genetic distance among 4 populations of S. chirtensis is 0.075 9 ± 0.030 2. A certain extent genetic variation among the populations had emerged and it maybe caused by the geographical isolation.
