摘要:[目的]白蜡是重要的工业原料,由白蜡虫雄虫在二龄若虫期分泌,该时期的长短对白蜡虫泌蜡量有重要影响,揭示该时期的蛋白表达特点对了解白蜡虫泌蜡和发育特点具有重要意义。[方法]本研究采用蛋白质组学的方法对白蜡虫雄虫二龄若虫前期和后期的蛋白表达进行分析,并采用荧光定量PCR对乙酰辅酶A羧化酶基因进行了表达谱分析。[结果]结果表明前期特有的蛋白有600个,后期特有的蛋白有464个,前期上调表达的蛋白有1 721个,下调表达的蛋白有1 478个。差异蛋白富集在昆虫表皮蛋白、保幼激素结合蛋白、蜕皮激素激酶等家族,很多差异蛋白参与泛素介导的蛋白降解、氧化磷酸化、肌动蛋白细胞骨架调等。脂肪酸延伸、类固醇激素等相关蛋白形成互作网络。与白蜡虫雄虫二龄若虫发育相关模块中的蛋白,富集在精细胞发育、分化、繁殖、蜡酯合成、昆虫激素合成等通路。乙酰辅酶A羧化酶基因在雌虫和雄虫二龄若虫期表达量高,与脂肪酸合成以及脂肪积累有关。[结论]白蜡虫雄虫二龄若虫发育后期白蜡合成相关蛋白表达下调,相关激素信号通路对白蜡虫泌蜡和发育可能具有调控作用,该研究对于了解白蜡虫泌蜡特点以及泌蜡量变化规律具有重要意义。
[Purpose]Chinses white wax is an important industrial raw material,secreted by Ericerus pela during the second instar nymphal stage.The duration of this stage is crucial for determining wax yield.In-vestigating protein expression patter during this stage is essential for understanding the mechanism of wax secretion and the developmental characteristic.[Method]A proteomic method was employed in this study to analyze the proteins in early and late second instar male nymphs.In addition,the expression profile of the acetyl-CoA carboxylase gene was analyzed through quantitative real-time polymerase chain reaction.[Result]Proteomic analysis identified 600 special proteins in early-stage nymphs and 464 in late-stage nymphs.Compared to the late stage,1721 proteins were up-regulated and 1478 were down-regulated in the early stage.Differently expressed proteins were significantly enriched in domains of insect cuticle pro-tein,haemolymph juvenile hormone binding protein,ecdysteroid kinase.The proteins were involved in ubi-quitin mediated proteolysis,oxidative phosphorylation,regulation of actin cytoskeleton et al.Interaction networks were identified among proteins related to fatty acid elongation,steroid hormone biosynthesis,and other metabolic processes.WGCNA module related to the development of male second instar nymph were involved in spermatid development,spermatid differentiation,reproduction,and wax biosynthesis,insect hormone biosynthesis.The ACC gene,which plays a central role in fatty acid synthesis and lipid accumu-lation,exhibited high expression levels in females and second-instar male nymphs.[Conclusion]The ex-pression of several key proteins involved in white wax biosynthesis declined during the late second-instar male stages.Hormone-related signaling pathways may regulate both wax secretion and nymphal develop-ment.These findings provide important insights into the molecular basis of wax production in E.pela and contribute to understanding the biological mechanisms underlying variation in wax yield.
