摘要:目的:考察银杏叶聚戊烯醇同系物以及其他15种类脂成分单体化合物对5种菌种(沙门菌、金黄色葡萄球菌、黑曲霉菌、大肠杆菌和枯草芽孢杆菌)的抑菌及协同作用。方法:正己烷作溶剂和空白对照,在样品质量浓度500 mg·L-1下,利用滤纸片法测定样品抑菌圈值;样品用正己烷配制成质量浓度分别为250,125,62.5,31.3,15.6,7.8,3.9 mg·L-1溶液并分散在培养基中,记录无菌生长样品浓度,以确定最小抑菌浓度(MIC)、最小杀细菌浓度(MBC)和最小杀真菌浓度(MFC)和计算分级抑菌浓度指数(FIC)方法来比较各样品对菌种的抑制作用。结果:橙花叔醇对4种细菌(沙门菌、金黄色葡萄球菌、大肠杆菌和枯草芽孢杆菌)的抑菌活性均为最强,抑菌圈范围为17.4~20.9 mm,MIC和MBC范围依次为3.9~15.6,31.3~62.5 mg·L-1;芳樟醇对黑曲霉菌的抑菌活性最强,抑菌圈为17.9 mm,MIC和MFC依次为7.8,31.3 mg·L-1;β-石竹烯与银杏叶聚戊烯醇混合样品组对枯草芽孢杆菌的协同抑制作用最强;当β-石竹烯与银杏叶聚戊烯醇质量比为43.42%∶56.58%时,其抗枯草芽孢杆菌协同作用最佳,此时测得实际值FIC指数为0.16,抑菌圈值为19.8 mm。结论:银杏叶类脂成分具有一定抑菌作用,部分类脂成分与聚戊烯醇混合具有协同抑菌效应。
Objective:Antibacterial/antifungal tests and synergistic interactions between polyprenols and other lipids(including fifteen known compounds) isolated from Ginkgo biloba leaves against five selected stains(Salmonella enterica,Staphylocococus aureus,Aspergillus niger,Escherichia coli and Bacillus subtilis) were carried out.Method:Antibacterial/antifungal actions and synergistic interactions were observed using a disc-diffusion method at 500 mg·L-1 in n-hexane(blank) by measuring inhibition halos and comparing minimum inhibitory concentrations(MIC),minimal bactericidal concentration(MBC),minimal fungicidal concentration(MFC) values by recording the sample concentration(250,125,62.5,31.3,15.6,7.8,3.9 mg·L-1 in n-hexane) as no bacteria growing and figuring out fractional inhibitory concentration(FIC) Index.Result:Nerolidol showed the highest activity against the four tapes of bacteria(S.enterica,S.aureus,E.coli and B.subtilis) among all the tested samples,and inhibition halos were 17.4-20.9 mm,MIC and MBC values were the range of 3.9-15.6,31.3-62.5 mg·L-1.linalool showed the highest activity against A.niger among all the tested samples and the GBP with β-caryophyllene mixture group had the strongest synergistic effect against B.subtilis among all mixture groups against the five tested strains,and inhibition halos were 17.9 mm,MIC and MFC values were 7.8,31.3 mg·L-1.The proportion of β-caryophyllene and GBP was 43.42% ∶56.58% that was determinated by Mixture Design as the optimal proportion of synergistic effect on GBP with isophytol against B.subtilis.In the meantime,the truth values of FIC index and inhibition halos were respectively 0.16,19.8 mm.Conclusion:G.biloba leaves polyprenols and other lipids have antimicrobial effects and the parts of lipid compounds with polyprenols have synergistic effects against the tested strains.
